Three luxury yachts have been destroyed and another is badly damaged after a massive blaze at a Sydney marina.

About 40 firefighters attended The Spit Marina in Mosman, on the Sydney Harbour, early on Friday and took more than two hours to extinguish the blaze.

Emergency services were initially called about 3.30am because one vessel had caught fire, before the flames spread to another three.

Four people had to be evacuated from the first yacht that caught fire, with one woman taken to hospital in a stable condition after being treated by paramedics for smoke inhalation.

Fire and Rescue NSW Superintendent Adam Dewberry said the three super yachts that were destroyed had all sunk, with attention turning to containing hazardous materials spilled as a result of the blaze.

"We have a fourth (yacht) that has suffered significant damage, that will have to be assessed, that is still floating," he said.

"We have installed booms around the debris site where the vessels have sunk to contain a lot of the material, including oils and fuels from those boats."

NSW Police said their initial inquiries revealed the fire is not being treated as suspicious.

The NSW Port Authority also attended the blaze, including in one of their boats.

The Spit Marina, located in the affluent harbour suburb of Mosman, describes itself as "located in one of Sydney Harbour's most prestigious boating precincts".
